Using thin layer chromatography to separate a mixture, one of the separated spots on the chromatogram.....?

...is still a mixture. What further step could be carried out to separate that spot into its components?Using thin layer chromatography to separate a mixture, one of the separated spots on the chromatogram.....?
Wait for the plate to dry completely. Turn the plate 90 degrees (previous spots at the bottom) and use a different carrier solvent/buffer with a different polarity/pH, it depends on what you are trying to separate. It is called 2D-TLC.
